Mechanism of Action

VIP binds VPAC1 and VPAC2 G-protein-coupled receptors to activate adenylyl cyclase and increase intracellular cAMP, producing vasodilation in pulmonary and systemic vasculature through smooth muscle relaxation. In immune cells, cAMP elevation suppresses NF-kB signaling, reducing pro-inflammatory cytokine production including TNF-alpha, IL-6, and IL-12 while promoting anti-inflammatory IL-10 and regulatory T-cell differentiation. VIP acts as a neurotransmitter and neuromodulator in the CNS, influencing circadian rhythm regulation through the suprachiasmatic nucleus, supporting neuronal survival through anti-apoptotic MAPK and PI3K/Akt signaling, and modulating autonomic tone. In the lung, VIP-mediated bronchial smooth muscle relaxation and mucosal secretion regulation contributes to airway patency and mucociliary defense.

Does VIP Nasal Spray require a prescription?

What are the possible side effects?

Mild transient flushing, nasal irritation, or headache may occur following administration due to VIP's vasodilatory activity.
