Niagen is nicotinamide riboside (NR), a bioavailable form of vitamin B3 that serves as a direct precursor to NAD+ (nicotinamide adenine dinucleotide). NAD+ is an essential coenzyme in every living cell, required for mitochondrial ATP production, DNA repair, and the activation of sirtuins, the longevity-associated protein deacetylases. Injectable delivery bypasses GI absorption variability to deliver therapeutic NR concentrations directly to plasma.

Mechanism of Action

NR enters cells via nucleoside transporters and is phosphorylated by NR kinases (NRK1/2) to form nicotinamide mononucleotide (NMN), which is then adenylated by NMNAT enzymes to produce NAD+. Elevated intracellular NAD+ activates SIRT1 and SIRT3 sirtuins, which regulate mitochondrial biogenesis, fatty acid oxidation, and genomic stability. NAD+ also serves as substrate for PARP enzymes involved in DNA strand break repair, and for CD38/CD157 ectoenzymes involved in calcium signaling and immune function. NAD+ declines significantly with age, and NR supplementation restores NAD+ pools in a dose-dependent manner.
