EGCG from green tea and vitamin E acetate provide layered free radical protection across aqueous and lipid phases. Caffeine contributes vasoactive and anti-inflammatory activity at the dermal level. The ointment base supports occlusive moisture retention and enhances penetration of co-actives through the stratum corneum.

Mechanism of Action

Glutathione inhibits tyrosinase by chelating copper at the active site and shifts melanin synthesis from eumelanin (dark) to phaeomelanin (lighter pigment), reducing skin darkening over time. It also directly quenches reactive oxygen species and regenerates vitamins C and E from their oxidized forms. EGCG (epigallocatechin gallate) is a potent polyphenol that scavenges free radicals, inhibits UV-induced NF-kB inflammatory signaling, and downregulates melanogenesis transcription factors including MITF. Vitamin E acetate (tocopheryl acetate) is converted to alpha-tocopherol in skin, where it provides lipid-phase antioxidant protection in cell membranes and reduces UV-induced lipid peroxidation. Caffeine inhibits phosphodiesterase to increase cAMP, supporting lipolysis in subcutaneous adipose and reducing puffiness. Topical caffeine also has documented anti-inflammatory activity and may inhibit UV-induced apoptosis in keratinocytes.
