L-Arginine

L-Arginine is a semi-essential amino acid that becomes conditionally essential under physiological stress, illness, and aging. It is the sole substrate for nitric oxide synthase (NOS) enzymes, making it the rate-limiting factor in endothelial nitric oxide production, vascular tone regulation, and tissue perfusion. Arginine also stimulates pituitary growth hormone release and is an obligate intermediate in the urea cycle. Injectable delivery provides consistent bioavailability independent of GI absorption or hepatic first-pass extraction.

Mechanism of Action

Arginine is converted to nitric oxide and citrulline by endothelial NOS (eNOS), inducible NOS (iNOS), and neuronal NOS (nNOS). Endothelial NO activates soluble guanylate cyclase in vascular smooth muscle, increasing cGMP and inducing vasodilation to improve tissue perfusion. Arginine stimulates growth hormone secretion from the anterior pituitary through inhibition of hypothalamic somatostatin release, increasing GH pulse amplitude. In the urea cycle, arginine is cleaved by arginase to produce ornithine and urea, supporting ammonia detoxification and downstream polyamine and proline synthesis. Arginine also activates mTORC1 signaling via the Ragulator-Rag GTPase complex, stimulating protein synthesis in anabolic tissue.

Lysine supplementation at high doses may reduce arginine cellular uptake through shared CAT transporter competition
