Overview

Amlexanox 40 mg / Tetradecylthioacetic Acid 200 mg

This compounded capsule combines two mechanistically distinct and complementary metabolic actives. Tetradecylthioacetic Acid (TTA) is a modified omega-3 fatty acid and PPAR-alpha pan-agonist that drives fat clearance from the bloodstream, promotes fatty acid oxidation in muscle and liver, and exerts cardioprotective and anti-inflammatory effects. Amlexanox is an IKKe and TBK1 kinase inhibitor originally developed as an anti-inflammatory and anti-allergic agent, repurposed for its ability to reverse obesity-induced suppression of AMPK, the master regulator of cellular energy expenditure.

Mechanism of Action

TTA: PPAR-alpha/delta/gamma pan-agonism upregulates fatty acid transport and beta-oxidation enzymes, shifts metabolism toward fat utilization, clears LDL, redistributes cholesterol to large HDL, and improves insulin sensitivity. Non-metabolizable structure ensures persistent PPAR activation.

Amlexanox: Inhibits IKKe and TBK1, relieving obesity-driven TBK1 suppression of AMPK to restore cellular energy expenditure. IKKe inhibition reduces NF-kB inflammatory signaling in adipose tissue, lowering the chronic inflammation that perpetuates insulin resistance.

Benefits

Fat Oxidation

TTA's PPAR-alpha pan-agonism upregulates fatty acid transport and beta-oxidation enzyme expression, promoting fat burning in liver and muscle while reducing adipose tissue mass.

LDL and Lipid Reduction

TTA clears lipoproteins from circulation, reduces LDL cholesterol, redistributes plasma cholesterol toward large cardioprotective HDL particles, and lowers blood pressure through vascular PPAR-delta activation.

AMPK Restoration

Amlexanox inhibits TBK1-mediated AMPK suppression to restore cellular energy expenditure capacity that obesity-driven inflammation has disabled.

Insulin Sensitivity

Dual-mechanism improvement through TTA's PPAR-gamma activity improving peripheral glucose uptake and amlexanox's IKKe inhibition reducing inflammatory insulin resistance.

Cardioprotection

TTA combines LDL reduction, blood pressure lowering, antioxidant defense, and anti-inflammatory activity for a multi-pathway cardioprotective profile.

Hemoglobin A1c

Reduction in HbA1c and fructosamine observed in preclinical models through the combined improvement in insulin sensitivity and fasting glucose achieved by both actives.

FAQs

Title or Question

Amlexanox Tetradecylthioacetic is a prescription therapy used for metabolic support.

Who should use Amlexanox Tetradecylthioacetic?

Patients seeking support for metabolic support may qualify after evaluation by a licensed provider.

Does Amlexanox / TTA require a prescription?

Yes. This is a compounded prescription preparation requiring a valid prescription from a licensed provider. Lab evaluation of lipids, glucose, and liver function is recommended at baseline.

What are the possible side effects?

TTA is generally well tolerated; mild GI discomfort, nausea, or loose stools may occur at initiation. Amlexanox at therapeutic oral doses may cause mild nausea, diarrhea, or rash in some patients. Liver function monitoring is appropriate with extended use given hepatic PPAR activation.

Who should not use Amlexanox / TTA ?

Not for patients with active hepatic disease or known hypersensitivity to either active. Patients with coagulation disorders should discuss TTA's omega-3 antiplatelet activity with their provider. Use during pregnancy or breastfeeding requires provider evaluation. Not established for patients under 18.

Are there any drug interactions?

TTA's PPAR-alpha activation may increase the metabolism of CYP4A-dependent substrates. Concurrent anticoagulants or antiplatelet agents require monitoring given omega-3 TTA's platelet-modulating activity. Amlexanox may interact with other anti-inflammatory or immunomodulatory agents; disclose all medications to your provider.

Is Amlexanox / TTA safe if I have a pre-existing medical condition?

Patients with hepatic impairment, coagulation disorders, or cardiovascular disease require individualized provider evaluation before starting. Diabetic patients on insulin or oral hypoglycemics should monitor blood glucose given the insulin-sensitizing effects of both actives. Periodic lipid panel and liver function monitoring is recommended during treatment.
