Selective Estrogen Receptor Modulator

Trans-isomer of clomiphene citrate. It selectively blocks estrogen receptors in the hypothalamus and pituitary, removing negative feedback on the HPG axis.

Mechanism of Action

Blocking estrogen receptors increases GnRH pulsatility, which stimulates pituitary release of LH and FSH. LH drives testicular testosterone production. FSH maintains sperm production.

Does Enclomiphene require a prescription?

Yes. Enclomiphene is a compounded prescription medication requiring a provider evaluation.

What are the possible side effects?

Possible side effects include mood changes, headaches, hot flashes, nausea, and elevated estradiol secondary to rising testosterone levels. Visual disturbances have been reported — discontinue use and contact your provider immediately if vision changes occur. Thromboembolic events have been reported rarely. Regular lab monitoring, including testosterone and estradiol, is recommended throughout treatment.

Who should not use Enclomiphene?

Not appropriate for men with primary hypogonadism (testicular failure), liver disease, or known hypersensitivity to clomiphene derivatives. Do not take if pregnant or breastfeeding.

Are there any drug interactions?

May interact with estrogen-containing medications and certain anticoagulants. Disclose all current medications to your provider.
